参加美国科研的体验

本文作者M同学,来自北京理工大学,机械工程专业,大学二年级,成绩优异,计划本科毕业后赴美读研究生,考虑到以后的就业情况,希望攻读生物医学工程相关专业研究生,转专业申请研究生,相比于本专业而言有一些困难,为了获得名校的录取,增加申请竞争力,为此报名了美国名校科研生物医学工程专业,参与选拔。参加美国名校科研,增加学术背景,开拓视野,获得真知。

选择在大二结束后的暑假参与科研,有独特的优势,学生表现优异,就成为了教授课题组的一员,以后大三、大四可以继续深入进行科研,这样会获得连续性的科研履历,更有利于美国顶尖大学研究生院的录取。

科研总结

本文是学生在美国大学科研学习结束后所写的感受,并在文后附录了学生的推荐信,供学生及家长参考。

Week 1 第一周

This week is the first I am working on a research project at American University. In the first week, I learned many basic skills in the lab. I learned how to use the pipette correctly. I thought it was very easy at first because I’ve used in my senior high school, but when I use it I found I was wrong. It needs to be accurate and it is very difficult especially use the pipette to have 1ul solution. I even got 0.5ul by mistakes. By trying it for lots of times, I can use it much well. Then I learn to mix a buffer A for HPLC. I know the composition of it and calculate the mass of each one the I dissolve each and use the PH detector to adjust the PH of it to 7. It’s the first time I have to make a reagent and it helps me know we need to think thoroughly about how much of each component should we add.

Then came the second day. I make the standard mixture for the HPLC. As I got the instruction, I think it’s a quite difficult one. First, I need to dissolve all the compounds into 2ml stock solution. I need to consider the purity of each compound. It really needs my patience because I need to fetch only 2mg and it needs to be very accurate. After I finished it, I need to calculate the volume needed for each one to make the mixture. I used my pen and calculate by hand at first, but my professor taught me to use excel to do the calculation, I soon learned how to do it and found it much more convenient. In the end, I adjust pH around 7 with pH paper, and that’s an easy task.

Before the third day, I used Google to search how does the HPLC work. And I know the function of each part of HPLC. One that day, I try to operate the HPLC. It was a little nervous because I always heard people saying that HPLC is difficult. However, when my professor gave me the guidance of it and show me how to do that, it became much easier. I follow the guidance and prime the pumps and then purge them using methanol, water and then buffers. Then I run the However standard mixture I made yesterday for the HPLC to analyze it. I firstly used the 50ul standard mixture. But after I had my lunch, there is something wrong with the computer and the data loss. I had to do it again and wait for 2 hours again. That’s frustrating. Finally, I successfully got the result below. Then I try 100ul as well then go the same result. It means that my standard mixture is the correct one.

I feel really excited about it’s the first time that I operate HPLC. Through the first week, I find it’s very interesting to do a project in the lab. I can learn much more things than in traditional classes. And I also know I should be more patient. What’s more, the most important thing I learned from the first week is the powerful function of excel. And I will learn to use excel more fluently in my daily study.

我感到非常兴奋,因为这是我第一次操作高效液相色谱。在第一个星期里,我发现在实验室里做一个项目很有趣。比起传统的课程,我能学到更多的东西。我也知道我应该更有耐心。而且,我从第一周学到的最重要的是Excel的强大功能。我将学会在日常学习中更流利地使用Excel。

Week 2 第二周

This week is the second week of doing research at American University.This week I continued my HPLC job. Things have been much more similar to me than the first week. Everything turns great this week. On the first day, I finish my first sample and protein. I followed the instruction provided by my professor. I firstly made some preparation such as having the tubes ready and preparing other compounds. Then I completed the sample1 step by step with the help of my professor. It’s the first time for me to see liquid nitrogen, and that’s really fantastic. Meanwhile, I also did the 20ul and 100ul injection of my standard mixture. Then on the second day, I continued my HPLC project. I injected 40ul and 60ul standard mixture. After several days’ practice, I can operate the machine much faster. At the same time, I made two samples of the tissue. It’s the major task I could do in this project. It’s quite difficult for me because I need to use the liquid nitrogen and I need to be really patient for it has some danger. Finally, I made it following the instruction. I am really delighted as I finish the task.

As the last day for this week came, I carried on my job for making samples 4,5,6,7 of the two groups. Meanwhile, I do my injections one by one. It becomes much easier to finish my job than I firstly finish the task. All the result seem great. In the end, I finished my job faster than we have thought. Therefore, the professor taught me how to analyse the data of the HPLC.

当这周的最后一天到来时,我继续我的工作,在两组中制作4,5,6,7个样品。同时我一个接一个地注射。完成工作比我先完成任务容易得多。所有的结果都很好。最后,我比我们想象的更快地完成了我的工作。因此,教授教我如何分析高效液相色谱数据。

In this week the operation in the experiments became much and much easier for me. Because I know each step of the experiments. For example, when I finish the samples, all the steps are same for each sample. As I have finished more and more samples, I can behave much better and much faster. But I also know that speed is not what we need in the end, we need the accuracy. One standard mixture’s result seems strange after the injection. I think I may make some mistakes in the process. From this I realize that we should spend more time considering about how to make the correct result instead of how to do the experiments faster. I will mind that in my study in the future.

Week3 第三周

This week is the third week I have been studying at American University. This week, in general, I finish preparing the samples, continue injecting the samples and starting the Lowry Protein assay. Although something was not so satisfactory, I finally finish all the tasks. The first day is another day for making and injecting samples. I tried to operate the machine without the instruction to check whether I have mastered the steps correctly. At first, it bothers me a lot, I need to recall what I did last time and double-check it to ensure I don’t make a mistake. Gradually it turns much easier for me as I carry on my experiments. And everything shows great in the end. I make another 4 samples and inject 3 of them. Meanwhile, I also make some data analyses in my spare time. I think I master the skills well after two-weeks practice.

On the second day I make the last three samples and finally finish my task for finishing 14 samples. I’m really proud of that because it’s my first time to do things like that. I find a lot of interests during I make those. Then I continue my injections as usual. Meanwhile, I finish my data analyses of the standard mixture. However, there is something wrong with that. Most of my standard mixtures only show 8 peaks instead of 9. It really confused me a lot. I dissolved 9 compounds in my standard mixture but why only show 8 peaks. Then my professor helps me with that. We check the nine compounds I made in the first day. And we find that the reason is that my Adenosine probably doesn’t dissolve in the resolution, that’s the missing peak. I think I might use the pure NH4OH to dissolve it. Then professor gives me his Adenosine data for me to calculate. I will put this in my mind. And I finally know the reason professor asked me to keep the each solution of every compounds.

Week 4 第四周

This week is the third week I have been studying at American University. And I must say time passed so quickly and I really enjoy this experience because I think I like using my hands to do some experiments. And this week I finished all my sample injections and analyzed all the data and make the tables of them.

本周是我在美国大学学习的第三周。而且我必须说时间过得如此之快,我真的很喜欢这种体验,因为我觉得我喜欢用手来做一些实验。本周我完成了所有样品注射并分析了所有数据并制作了表格。

On the first day, A good news for today is that the HPLC’s problem has been solved, so I carry on my injections. And I have finished the last four samples and successfully get the result. Then comes the data analyses. Professor introduces a new software called Graphpad prism to me. That is really powerful and can produce the curves according to the data. I soon learn to operate it. I’m really glad to master another useful tool for analyzing the data.

在第一天,今天的好消息是HPLC的问题已经解决,所以我继续我的注射。我已完成最后四个样本并成功获得结果。然后是数据分析。教授向我介绍了一款名为Graphpad prism的新软件。这非常强大,可以根据数据生成曲线。我很快学会了操作它。我很高兴掌握另一种分析数据的有用工具。

Then came the second day and sadly my last day for experiments. Professor He then introduced the method to figure out each peak stands for which composition. I soon got it and started to analyse all the samples. After one-morning’s effort, I finally got all the data in an excel. And professor showed me how to use Graphpad prism to deal with the data. And then I got the graphs of them. However, something wrong happened. Some of my data failed to show the correct trend and much larger than others. As I looked back on my experiment, I thought there was something wrong with my protein assay. It was really frustrated and disappointed. But professor He told me on the first day that error is not a bad thing and you should learn from the error. I think this is very meaningful and I think I should be more patient to do anything so that I could make less mistakes.

Then my internship ends. I think I do gained a lot from this experience. I know the basic process and methods of research in the US and I realize how precise is required in any research, which need to be really patient. Also, I have made a lot of friends during this experience and know a lot about city. What’s most important to me is that I have know more about Medical area and I can have more choices when I decide my major in the future after graduation.

I really enjoy this experience. I’m very grateful for professor He to give me a chance to have this experience and give me very patient instruction.

First, I have been repeatedly impressed by his outstanding research ability. He was both driven and teachable, willing to invest considerable time and energy into his work and open to receive instructions to refine his skills. He is a quick learner, able to readily assimilate the goals and methods of the tasks he was assigned to. For example, within a short period, he mastered the correct usage of the pipette, learned to prepare a solution named buffer A for HPLC, familiarized himself with the functions of HPLC, and acquired the skills of processing data with Excel and Graphpad prism. Besides, he applied himself diligently and with enthusiasm throughout the whole research program by flexibly practicing what he had gained in the first stage of our research. I believe that he has made significant progress at American University, and the final report with logical structure and vivid graphs was a strong proof of his harvest.

Second, I appreciate his positive attitude towards research. It has been clear to me that he enjoys research a great deal. He often used the word “interesting” to describe his experiment, and considered it an awesome experience when firstly applied liquid nitrogen in his experiment. When encountered with problems during the research process, he never complained; instead, he positively attempted to get through it by himself first and provided multiple solutions. Later, he would discuss with me and revise his solutions accordingly. He thought mistakes were not bad, while lessons drawn from mistakes were most valuable. I think such a proactive research attitude is the cornerstone for him to be an excellent researcher in the near future.

第二,我欣赏他对研究的积极态度。我很清楚他很喜欢研究。他经常用“有趣”这个词来描述他的实验,并认为这是一个了不起的经验时,首次应用液氮在他的实验。在研究过程中遇到问题时,他从不抱怨;相反,他积极尝试自己先解决问题,并提供了多种解决方案。稍后,他会和我讨论并相应地修改他的解决方案。他认为错误并不坏,而从错误中吸取的教训是最有价值的。我认为这种积极主动的研究态度是他在不久的将来成为一名优秀研究员的。
